Continuously Screening Biofilms in Real Time

xCELLigence impedance-based real-time analyzers are successfully applied in many areas of adherent eukaryotic cell research.

In a new article, published recently in PLoS ONE, Diana Gutierrez et al. describe how they validated this method for monitoring the formation of single-species biofilms of three bacteria (Staphylococcus aureus, Staphylococcus epidermidis and Streptococcus
mutans) in real time.

xCELLigence assays allowed the distinction between biofilm-producers and non-producers (S. aureus & S. epidermis) and monitoring of biofilm formation of S. mutans when sucrose was present in the medium. Also, impedance values allowed for discrimination among the biofilm-producing strains.

Antimicrobial treatment with a bacteriophage was continuously monitored and treatment with bacteriophage endolysin (LysH5) led to the monitoring of the removal of a preformed biofilm.

The authors conclude that xCELLigence technology opens new encouraging opportunities in the biofilm-related research arena, being a technology that can be easily used, is fast and reliable for screening of biofilm-forming strains as well as for evaluating bio-actives that interfere the biofilm formation.
The method is not limited by an endpoint measurement since it allows continuous monitoring and avoids staining procedures being a label-free technique.
